Addition of fertilizers, pesticides and industrial waste also makes water unsafe but this is not so much a problem in hills, as in the adjoining plains having SEZ´s. In hills which are densely populated, sewage, rains and flood continues to be major a problem. From the practical stand point determination of microbial load and identification of method remains to most important to all sell water quality. At Samrpit Sewa Sansthan,we have lab facility to check water quality. Most provable number (MPN) method is applied. The method is also known as multiple tube technique. It commonly employs five tubes and other three dilutions for estimating MPN of coliform or other indicator organism per 100ml. The principle of coli form enumeration by MPN/100ml method is that presence of coli form in water sample can be demonstrated at 370C in 48 hr by production of gass following incubations of such sample in Mac Conkry broth 10, 1 and 0.01ml portion of water is incubated in five broth tubes each containing an inverted Durham´s tube tarp gass. The 10ml portion of water is the incubated in 10ml of double Mac Conkry and 0.1ml portion and incubated in 5ml of single distance Mac Conkry.
